It binds to both DNA and RNA in a sequence-independent manner and with a >30-fold fluorescence enhancement. The DNA binding of each Ethidium Homodimer covers four base pairs and is believed to occur by intercalation. Because the dye is highly positively charged, it cannot cross cell membranes to stain living cells. It is useful for detecting nucleic acids in solution, or for selectively staining dead cells with damaged plasma membranes.

The CERTISTAIN® dry dye Orange G (C.I. 16230) is used for the preparation of a staining solution for Papanicolaou’s staining in cytology in material of human origin. The cytological staining technique developed by Papanicolaou is still the standard method in cancer and hormone cycle diagnosis. The effect of the Orange G staining solution is considerable in smears with keratinized cells under acidic pH conditions, when the obvious orangeophilic cytoplasmic stain is recognizable and may point to the presence of hyperkeratosis, HPV infection or carcinoma cells.
